Think about your day for a second.

You woke up, felt fresh air on your face as you walked out the door,

you probably don't think about it very often at all.

And that's that all those sensations, feelings, decisions and actions

are mediated by the computer in your head

called your brain.

Now, the brain may not look like much from the outside --

a couple pounds of pinkish-gray flesh,

amorphous.

But the last 100 years of neuroscience have allowed us to zoom in on the brain

and to see the intricacy of what lies within.

And they've told us that this brain is an incredibly complicated circuit

made out of hundreds of billions of cells called neurons.

Now, unlike a human-designed computer,

where there's a fairly small number of different parts,

and we know how they work because we humans designed them,

the brain is made out of thousands of different kinds of cells,

maybe tens of thousands.

They come in different shapes; they're made out of different molecules;

Let's make it concrete.

There's a class of cells,

a fairly small cell, an inhibitory cell, that quiets its neighbors.

It's one of the cells that seems to be atrophied

in disorders like schizophrenia.

It's called the basket cell.

And this cell is one of the thousands of kinds of cell

that we're learning about.

New ones are being discovered every day.

As just a second example:

these pyramidal cells, large cells, can span a significant fraction

They're excitatory.

And these are some of the cells that might be overactive

Every one of these cells is an incredible electrical device.

They receive inputs from thousands of upstream partners

and compute their own electrical outputs,

which then, if they pass a certain threshold,

And this process, which takes just a millisecond or so,

happens thousands of times a minute in every one of your 100 billion cells,

as long as you live and think and feel.

So how are we going to figure out what this circuit does?

Ideally, we could go through this circuit

and turn these different kinds of cell on and off

and see whether we could figure out

which ones contribute to certain functions

and which ones go wrong in certain pathologies.

وماهي تلك التي يتقوض عملها ضمن ظروف معينة

فنحن ان استطعنا ان نحفز الخلايا .. يمكننا ان نرى ماهية الطاقة الناجمة عنها

If we could activate cells, we could see what powers they can unleash,

وعلى ماذا تؤثر .. وماهو التي تحفزه .. او تقوضه

what they can initiate and sustain.

وان استطعنا ان نطفىء بعض الخلايا

If we could turn them off,

then we could try and figure out what they're necessary for.

يمكننا ان نعرف لم هي موجودة .. وماهي اهميتها

والقصة التي اريد ان اخبركم بها اليوم

And that's the story I'm going to tell you about today.

هي خلاصة ما كنا نقوم به على مدى ال11 عاما الماضية

And honestly, where we've gone through over the last 11 years,

نحاول ان نجد طرق

through an attempt to find ways

of turning circuits and cells and parts and pathways of the brain

لكي نشغل الدارات و السيالات العصبية و الموصلات الكهربائية في الدماغ

ونطفؤها

on and off,

لكي نستوعب آلية عملها العلمية

both to understand the science

and also to confront some of the issues that face us all as humans.

ولكي نحاول ان نحل بعض المشاكل

التي تواجه البشرية

Now, before I tell you about the technology,

وقبل ان اخبركم عن هذه التكنولوجيا

the bad news is that a significant fraction of us in this room,

فان الخبر السيء الذي اريد ان اقوله لكم .. ان عدد كبير ممن هو موجود في هذه الغرفة

ان استطاع أن يعيش عمراً مديداً

if we live long enough,

will encounter, perhaps, a brain disorder.

سوف يوجه نوعا من الاعطاب الدماغية

وحتى اليوم مليار شخص

Already, a billion people have had some kind of brain disorder

يعاني اليوم من عطب دماغي

يقوض عملهم وحياتهم

that incapacitates them.

The numbers don't do it justice, though.

والارقام تلك لا تشمل الاعداد الحقيقية

These disorders -- schizophrenia, Alzheimer's,

للفصام والزهايمر

والاحباط والادمان

depression, addiction --

they not only steal away our time to live,

هذه الخلايا .. لا تتحكم بنا فحسب .. بل تغيرنا

they change who we are.

They take our identity and change our emotions

وهي تتحكم بعواطفنا و شخصيتنا و إنطباعاتنا

and change who we are as people.

وهي تغير ماهيتنا كبشر

Now, in the 20th century,

وفي القرن ال20

تولف لدينا أمل

there was some hope that was generated

through the development of pharmaceuticals for treating brain disorders.

عبر تطوير عدة مواد كيميائية استطاعت ان تشفي البعض من الاعتلالات الدماغية

ورغم تطوير العديد من العقاقير الطبية

And while many drugs have been developed

that can alleviate symptoms of brain disorders,

التي يمكنها ان توقف بعض الاعراض الناتجة عن الاعتلالات الدماغية

لا يوجد اي واحد منها يمكن اعتباره علاج

practically none of them can be considered to be cured.

In part, that's because, if you think about it,

لان ما نقوم به هو تعويم الدماغ بمواد كيميائية

we're bathing the brain in a chemical --

وعليه فان العديد من الخلايا " الدارات الكهربائية "

this elaborate circuit, made of thousands of different kinds of cell --

المصنوعة من الآلاف من الخلايا الرديفة

تتأثر بذلك العلاج " العقار الكيميائي " على نحو سواء

is being bathed in a substance.

That's also why most of the drugs, not all, on the market

ولهذا السبب الكثير من العقاقير الموجودة في السوق

تؤدي الى العديد من الاعراض الجانبية

can present some kind of serious side effect too.

ولكن هناك بعض الاشخاص الذين حصلوا على " نتائج مرضية "

Now some people have gotten some solace from electrical stimulators

جراء محفزات كهربائية وضعت في الدماغ

that are implanted in the brain,

for Parkinson's disease or cochlear implants.

من اجل إعتلال باركنسون

ان زراعة المحفزات الكهربائية

These have indeed been able to bring some kind of remedy

كانت حتما

نوع من الحل المرضي

to people with certain kinds of disorders.

اشخاص ذوي الاعتلالات

But electricity also will go in all directions --

ولكن الكهرباء كانت تنتشر في كل الاتجاهات

the path of least resistance --

وخاصة تلك ذات المقاومة القليلة

which is where that phrase, in part, comes from,

والذي هو لب المشكلة في تلك الحلول

and will also affect normal circuits,

وهذا يؤثر ايضاً على الدارات الكهربائية الغير معتلة وتلك التي يراد اصلاحها ..

as well as the abnormal ones you want to fix.

لذا .. نحن نعود مرة اخرى الى المشكلة ذاتها " تعويم الحل على الدماغ كله "

So again, we're sent back to the idea of ultraprecise control:

والمطلوب التحكم الدقيق جداً بالخلايا المعطوبة

Could we dial in information precisely where we want it to go?

فهل نستطيع ان نضبط عملية حركة السيالات الكهربائية بصورة دقيقة ؟

حسناً .. لقد بدأت في مجال علم الاعصاب منذ نحو 11 سنة

So, when I started in neuroscience 11 years ago --

I had trained as an electrical engineer and a physicist --

لقد تدربت على أن اكون مهندس كهرباء و طبيب

the first thing I thought about was,

وكان الامر الذي بدأت في العمل عليه

if these neurons are electrical devices,

هو اعتبار ان هذه الاعصاب ليست سوى ادوات كهربائية

all we need to do is to find some way of driving those electrical changes

وكل ما نريده هو ايجاد طريقة

لتحفيز وتغير هذه الدارات الكهربائية من على بعد

at a distance.

وان استطعنا ان نحفز او نشغل خلية واحدة

If we could turn on the electricity in one cell but not its neighbors,

دون ان نقوم بالامر ذاته لمحيطها من الخلايا

that'd give us the tool to activate and shut down these different cells

سوف نستطيع ان نجد طريقة لكي نتحكم بتفعيل وتعطيل الخلايا المختلفة

ويمكننا ان نكتشف جراء ذلك كيفية تواصلها ومشاركتها

to figure out what they do

and how they contribute to the networks in which they're embedded.

في الشبكات الكهربائية التي تحويها

ويمكننا ايضاً ان نضبط عملية الدفق الكهربائي كما نريد

It would also allow us to have the ultraprecise control we need

لكي نصلح عمليات الخلايا العصبية واعتلالاتها

to fix the circuit computations that have gone awry.

التي تخرج منها على الدوام

Now, how are we going to do that?

ولكن كيف يمكننا القيام بذلك ؟

Well, there are many molecules that exist in nature

حسناً .. في الحقيقة هناك الكثير من الجزيئات في الطبيعة

التي تستطيع ان تحول الضوء الى كهرباء

which are able to convert light into electricity.

يمكن ان تتصوروهم كأنهم بروتينات صغيرة

You can think of them as little proteins that are like solar cells.

تعمل كخلايا كهروضوئية

If we install these molecules in neurons somehow,

وان استطعنا ان نقوم بتركيب هذه الجزيئات على الاعصاب بطريقة ما

then these neurons would become electrically drivable with light,

فان هذه الخلايا العصبية يمكن ان تحفز بواسطة الضوء عن طريق تلك الجزيئات

and their neighbors, which don't have this molecule, would not.

بينما لن تحفز الخلايا العصبية التي تقع بقربها والتي لا تملك تلك الجزيئات

There's one other magic trick you need to make this happen:

ويمكن القيام بذلك بحركة سحرية واحدة

the ability to get light into the brain.

The brain doesn't feel pain.

Taking advantage of all the effort

that's gone into the internet, telecommunications, etc.,

you can put optical fibers connected to lasers

to activate -- in animal models, for example, in preclinical studies --

these neurons and see what they do.

لاجل تحفيز الاعصاب ومعرفة تفاعلها ..

والآن كيف يمكننا القيام بذلك ؟

So how do we do this?

Around 2004, in collaboration with Georg Nagel and Karl Deisseroth,

حوالي عام 2004

وبالتعاون مع "جيرهارد ناجيل " و " كار ديسيروث "

this vision came to fruition.

اصبحت تلك الفكرة واقعاً

There's a certain alga that swims in the wild,

هناك نوع من الطحالب يعيش في البحار

and it needs to navigate towards light in order to photosynthesize optimally.

يبحث عن الضوء ويتحرك نحوه

ولكي يستطيع ان يحدد موقع الضوء و يميزه

And it senses light with a little eyespot,

يقوم بذلك باستخدام مجس ضوئي

which works not unlike how our eye works.

يعمل بصورة مختلفة عن عين الانسان

In its membrane, or its boundary,

انها غشاء.. او رقاق

it contains little proteins

يحوي بروتينات

تستطيع تحويل الضوء الى سيالات كهربائية

that indeed can convert light into electricity.

وهذه الجزئيات التي تقوم بذلك تدعى "channelrhodopsins"

These molecules are called channelrhodopsins.

And each of these proteins acts just like that solar cell

وكل بروتين من تلك يعمل كخلية كهروضوئية

that I told you about.

وعندما يترطم الضوءالازرق بها . فان بوابة تفتح

When blue light hits it,

it opens a little hole and allows charged particles to enter the eyespot;

فتتحرر الجزيئات التي تدخل المجس الضوئي

والذي يقوم باستخدامها لتوليد شحنة كهربائية

that allows this eyespot to have an electrical signal,

كما تماما في الخلية الكهروضوئية التي تقوم بشحن البطاريات

just like a solar cell charging a battery.

So what we need to do is take these molecules

وما نريد القيام به .. هو ان نأخذ تلك الجزئيات

ونقوم بصورة ما بتركيبها على الخلايا العصبية

and somehow install them in neurons.

And because it's a protein,

ولانها بروتينات في الاساس - تلك الجزئيات -

it's encoded for in the DNA of this organism.

فانها تحوي الحمض النووي في تركيبتها

So all we've got to do is take that DNA,

لذا فكل ما نحتاج اليه هو ان ناخذ هذا الحمض النووي

put it into a gene therapy vector, like a virus,

ونقوم بتعديله مخبريا لكي نحوله الى شبيه الفيروسات

and put it into neurons.

نضعه على الخلايا العصبية

And this was a very productive time in gene therapy,

ومن ثم بتكاثره وهذا يحدث بصورة سريعة

and lots of viruses were coming along,

سيخرج الكثير من اشباه الفيروسات الخاصة بنا وتنتشر في الخلايا العصبية المستهدفة

so this turned out to be fairly simple.

وقد تبين ان القيام بذلك ليس بهذه الصعوبة

Early in the morning one day in the summer of 2004,

we gave it a try, and it worked on the first try.

قمنا بتجربة أولية .. ونجحنا فيها

You take this DNA and put it into the neuron.

The neuron uses its natural protein-making machinery

فيستخدمه العصبون كأداة لصناعة البروتين

to fabricate these little light-sensitive proteins

ويقوم بتشكيل بروتينات ذات المجسات الضوئية

وتُنشر على تلك الخلايا

and install them all over the cell, like putting solar panels on a roof.

بنفس طريقة وضع الألواح الكهروضوئية على أسطحة المنزل

And the next thing you know,

وإذا بنا نحصل الآن على

you have a neuron which can be activated with light.

عصبون يمكن ان يتم تفعيله بواسطة الضوء

وهذا تقدم كبير جداً

So this is very powerful.

One of the tricks you have to do is figure out how to deliver these genes

وكانت احدى المعضلات التي واجهتنا

هي كيفية توزيع تلك البروتينات على نوع معين من الخلايا

to the cells you want and not all the other neighbors.

دون الخلايا المجاورة

واستطعنا القيام بذلك عن طريق برمجة الفيروسات

And you can do that;

you can tweak the viruses so they hit some cells and not others.

بحيث تهاجم نوع من الخلايا دون الاخرى

وهي وسيلة تمكننا من

And there's other genetic tricks you can play

صنع خلايا عصبية تتجاوب مع الضوء وتتفاعل معه

in order to get light-activated cells.

ان هذا المجال \ حقل العمل .. بات يعرف اليوم ب علم الجينات الضوئية

This field has now come to be known as "optogenetics."

And just as one example of the kind of thing you can do,

واحد الاشياء التي يمكننا القيام بها عن طريق تلك التقنية

هو اننا ندخل الى شبكة عصبية معقدة

you can take a complex network,

use one of these viruses to deliver the gene

ونستخدم هذه الفيروسات من اجل الوصول الى جينات معينة

محددة ضمن شبكة عصبية متشابكة ومعقدة وكثيفة

just to one kind of cell in this dense network.

And then when you shine light on the entire network,

ومن ثم عندما توجه ضوء مشع الى الشبكة تلك

just that cell type will be activated.

فان الخلايا المعنية سوف تُفعل

For example, let's consider that basket cell I told you about earlier,

لذا على سبيل المثال .. لنعد الى تلك الخلايا " السلال " التي اخبرتكم عنها فيما سبق

هذه التي تضمر في حال الفصام

the one that's atrophied in schizophrenia

وها هي المثبطة

and the one that is inhibitory.

ان استطعنا ان نوصل الجينات ( الضوئية ) الى تلك الخلايا

If we can deliver that gene to these cells --

والتي لن تعمل في حال عمل الخلايا وتفريغها للشحنات لانها تعمل بالضوء

they won't be altered by the expression of the gene, of course --

ولكن عندما نسلط ضوء ازرق على شبكة الدماغ العصبية

then flash blue light over the entire brain network,

just these cells are going to be driven.

فان تلك الخلايا سوف تفعل ان لم تكن مفعلة

وما ان نطفىء الضوء حتى يعود كل شيء الى وضعه الطبيعي

And when the light turns off, these cells go back to normal;

دون ان تقوم تلك الخلايا العصبية بطرد البروتينات او النفور منها

there don't seem to be adverse events.

Not only can you study what these cells do,

ويمكننا ان نستخدم هذه الطريقة لدراسة الخلايا

what their power is in computing in the brain,

والامر الاكثر اهمية هو معرفة آلية عمل الدماغ وخلاياه

كما يمكننا في حال ما

you can also use this to try to figure out

ان نقوم بتفعيل الخلايا ..

if we could jazz up the activity of these cells

ان كانت في حالة ضمور

if indeed, they're atrophied.

I want to tell you some short stories about how we're using this

والان اريد ان اخبركم قصتين قصيرتين

عن كيفية استخدامنا لتلك التقنية

both at the scientific clinical and preclinical levels.

وكلاهما تجارب مخبرية سريرية و غير سريرية

One of the questions that we've confronted is:

بداية حاولنا الاجابة على السؤال التالي

What signals in the brain mediate the sensation of reward?

ماهي الاشارات الكهربائية التي يولدها الدماغ من اجل الشعور بالانجاز

Because if you could find those,

وان استطعنا ان نقوم بذلك

those would be some of the signals that could drive learning;

يمكننا ان نحفز تلك الخلايا لكي ندفع في سبيل التعليم

لان الدماغ سوف يقوم بالاشياء التي ستشعره بالانجاز

the brain will do more of what got that reward.

ونفس الامر ينسحب على الادمان ..

These are also signals that go awry in disorders such as addiction.

فان استطعنا معرفة هذه الخلايا ..

So if we could figure out what cells they are, we could maybe find new targets

لربما أمكننا ان نجد أهداف أخرى

لنصمم عقاقير جديدة تستهدفها أو تصورها ..

for which drugs can be designed or screened against

أو ربما يمكننا ان نزرع اقطاب في اماكن معينة

or maybe places where electrodes could be put in

لكي نساعد هؤلاء الذين يعانون من اعاقات حادة

for people who have severe disability.

To do that, we came up with a very simple paradigm

وللقيام بذلك .. خرجنا بنموذج بسيط

بالتعاون مع مجموعة فيوريلا

in collaboration with the Fiorillo group,

هنا في احد اطراف هذا الصندوق الصغير

where, if the animal goes to one side of this little box,

ان ذهب الفأر الى هناك سوف يوجه نحوه ضوء أزرق

it gets a pulse of light.

من اجل ان يتم جعل الخلايا حساسة تجاه الضوء الازرق

And we'll make different cells in the brain sensitive to light.

وان استطاعت تلك الخلايا ان تولد الشعور بالرضى

If these cells can mediate reward, the animal should go there more and more.

فسوف يذهب الفأر الى تلك الجهة مرارا وتكرارا

وهذا ما حدث

And that's what happens.

The animal goes to the right-hand side and pokes his nose there

لقد ذهب الفأر الى الجهة اليمنى وقم بلكز الجدار بأنفه

وما ان يفعل ذلك حتى يحصل على ضوء ازرق في كل مرة قام بذلك

and gets a flash of blue light every time he does it.

وسوف يقوم بذلك مئات والالف من المرات

He'll do that hundreds of times.

هذه اعصاب الدوبامين

These are the dopamine neurons,

والتي ربما سمعتهم عنها والتي هي اصل الشعور بالمتعة والسعادة

in some of the pleasure centers in the brain.

We've shown that a brief activation of these is enough to drive learning.

Now we can generalize the idea.

Instead of one point in the brain, we can devise devices that span the brain,

that can deliver light into three-dimensional patterns --

عن طريق حزم من الالياف الضوئية

arrays of optical fibers,

each coupled to its own independent miniature light source.

وعليه يمكننا ان نقوم بأمور متراكبة فيما يتعلق بالجسم الحي - الدماغ -

Then we can try to do things in vivo

مثل الكشف اللحظي

that have only been done to date in a dish,

like high-throughput screening throughout the entire brain

ومثل الفرز الفائق الدقة للإشارات في جميع أنحاء الدماغ التي يمكن أن تسبب \ تنتج

بمهمات معينة او اوامر معينة

for the signals that can cause certain things to happen

ويمكننا ان نستخدم ذات الامر لاهداف طبية

or that could be good clinical targets for treating brain disorders.

One story I want to tell you about is:

How can we find targets for treating post-traumatic stress disorder,

والذي يعتبر شكل من أشكال القلق والخوف غير المنضبط.

a form of uncontrolled anxiety and fear?

واحد الامور التي نقوم بها

One of the things that we did was to adopt a very classical model of fear.

هو اننا اخذنا نقوم بالتجارب على نموذج خوف اعتيادي

This goes back to the Pavlovian days.

وهذا يعود الى أيام تجارب بافلوف

It's called Pavlovian fear conditioning, where a tone ends with a brief shock.

The shock isn't painful, but it's a little annoying.

انها ليست مؤلمة .. ولكنها مزعجة

وعلى مدى الزمن .. وفي هذه الحالة .. في حالة الفأر

And over time -- in this case, a mouse,

حيث يعتبر نموذج جيد .. والفأر يستخدم في هذه التجارب على طيف واسع

which is a good animal model, commonly used in such experiments --

يعتاد الفأر الخوف من تلكم الاومر او العبارات

the animal learns to fear the tone.

فيتجاوب معها بالجزع وعدم الحراك

It will react by freezing, sort of like a deer in the headlights.

كما يفعل الغزال حين مواجهة الاضواء العالية

Now the question is: What targets in the brain can we find

وكان السؤال المطروح حينها .. ماهي الخلايا التي

that allow us to overcome this fear?

يجب ان نستهدف من اجل التغلب على ذلك الخوف ؟

ان ما نفعله هو ان نغير " توجه " الحزم الضوئية

So we play that tone again, after it's been associated with fear.

حتى نجد تلك المتعلقة بالخوف

ويمكننا تفعيل عدة مناطق مختلفة في الدماغ

But we activate different targets in the brain,

باستخدام الحزم الضوئية .. التي اريتكم ايها فيما سبق

using that optical fiber array I showed on the previous slide,

لمعرفة اي تلك التي يجب ان تفصل

in order to try and figure out

which targets can cause the brain to overcome that memory of fear.

that we're working on now.

This is an area in the prefrontal cortex, a region where we can use cognition

to try to overcome aversive emotional states.

The animal hears a tone. A flash of light occurs.

There's no audio, but you see that the animal freezes --

ان هذا الصوت يعني له " الخوف والجزع " بحسب ذاكرته

the tone used to mean bad news.

There's a little clock in the lower left-hand corner.

You can see the animal is about two minutes into this.

And the same tone is going to play, and the light is going to flash again.

to overcome the expression of this fear memory.

على مدى العامين الماضيين .. عدنا عبر شجرة الحياة الى بداياتها

Over the last couple years, we've gone back to the tree of life,

من اجل ان نجد طريقة من اجل ان نوقف بعض الدارات الكهربائية في الدماغ

because we wanted to find ways to turn circuits in the brain off.

لاننا ان استطعنا ذلك .. فهذا سيكون تقدماً باهراً

If we could do that, this could be extremely powerful.

If you can delete cells for a few milliseconds or seconds,

وان استطعنا ان نوقف بعض الخلايا لاجزاء من الثانية

you can figure out what role they play in the circuits in which they're embedded.

فيمكننا على الفور معرفة الدور التي تقوم به هذه الخلايا

واي الدارات التي تتشابك معها

We surveyed organisms from all over the tree of life --

والان نحن نقوم باستبيان كل تلك المعلومات من خلال المنظومات الحيوية المتوزعة على شجرة الحياة

في كل مملكة من الممالك الحية .. دون الحيوانات .. ونحن نجد تمايز قليل

every kingdom of life but animals; we see slightly differently.

لقد اكتشفنا عدة انواع من الجزئيات مثل " halorhodopsins أو archaerhodopsins "

We found molecules called halorhodopsins or archaerhodopsins,

والتي تتجاوب مع اللون الاخضر والاصفر

that respond to green and yellow light.

And they do the opposite of the molecule I told you about before,

وهي تقوم بعكس الأمر الذي تقوم به تلك التي اخبرتكم عنها من ذي قبل

التي تتجاوب مع اللون الازرق

with the blue light activator, channelrhodopsin.

Let's give an example of where we think this is going to go.

دعوني اخبركم عن توقعاتنا لما يمكن ان نصل اليه جراء كل هذا

Consider, for example, a condition like epilepsy,

الآن تخيلوا حالة مثل الصرع

where the brain is overactive.

بحيث يكون فيها الدماغ " نشطاً " بشدة

Now, if drugs fail in epileptic treatment,

وان فشلت عقاقير الصرع في معالجة ذلك

one of the strategies is to remove part of the brain,

فان احدى طرق الحل تكمن في اجتثاث جزء من الدماغ

ولكن هذا الفعل لايمكن التراجع عنه لاحقا .. وله الكثير من الآثار الجانبية

but that's irreversible, and there could be side effects.

ولكن .. ماذا لو استطعنا ان نطفىء بضع خلايا من الدماغ لفترة وجيزة من الزمن

What if we could just turn off that brain for the brief amount of time

حتى تنتهي نوبة الصرع

until the seizure dies away,

and cause the brain to be restored to its initial state,

ويعود الدماغ الى حالته الطبيعية و وضعه الاعتيادي

like a dynamical system that's being coaxed down into a stable state?

كما أنظمة الضبط في المحركات الكبيرة التي تستخدم لحين استعادة الحالة الاعتيادية

هذا العرض يوضح لكم هذا المفهوم

This animation tries to explain this concept

where we made these cells sensitive to being turned off with light,

حيث تم الوصول الى طريقة لإطفاء هذه الخلايا الحساسة بواسطة الضوء

ما ان يسلط نحوها

and we beam light in,

and just for the time it takes to shut down a seizure,

لمدة تكفي حتى حين انتهاء النوبة

ونحن نأمل ان نستطيع القيام بذلك فعلاً

we're hoping to be able to turn it off.

وحتى اليوم لا معلومات اكيدة لدي لكي اعرضها عليكم

We don't have data to show you on this front,

وكلننا متفائلون جداً حيال ذلك ..

but we're very excited about this.

I want to close on one story, which we think is another possibility,

والان اريد ان انهي بقصة

وهي عن امكانية أخرى

which is that maybe these molecules, if you can do ultraprecise control,

والتي مكن ان يستخدم فيها هذه الجزئيات .. ان استطعنا ان نصل الى مستوى تحكم معين

يمكننا ان نستخدمها في الدماغ ذاته

can be used in the brain itself to make a new kind of prosthetic,

لكي نستطيع ان نشكل اجزاء جديدة .. اجزاء ضوئية " بصرية "

an optical prosthetic.

لقد اخبرتكم في البداية ان المحفزات الكهربائية شائعة

I already told you that electrical stimulators are not uncommon.

وان 75 الف شخص من المصابين بمرض باركنسون لديهم اجهزة مشابهة مزروعة

Seventy-five thousand people

have Parkinson's deep-brain stimulators implanted,

وربما 100000 لديهم قواقع مزروعة

maybe 100,000 people have cochlear implants, which allow them to hear.

تمكنهم من السمع بصورة افضل

ولكن ماذا لو استطعنا ان نرسل نوع من الجينات الى تلك الخلايا ..

Another thing -- you've got to get these genes into cells.

سيكون هذا علاج جيني يمكن ان يتطور

A new hope in gene therapy has been developed,

because viruses like the adeno-associated virus --

باستخدام اسلوب الفيروسات مثل فيروس " أدينو "

والذي معظم من في القاعة هنا مصاب به

which probably most of us around this room have;

ولكنه لا يحوي اية اعراض جانبية

it doesn't have any symptoms --

وهو يستخدم في علاج المئات من المرضى

have been used in hundreds of patients

to deliver genes into the brain or the body.

من اجل ايصال الجينات الى مناطق معينة من الجسد او الدماغ

And so far, there have not been serious adverse events

ولكن حتى اليوم لم يحدث تقدم كبير في هذا المجال

مرتبط بذلك الفيروس

associated with the virus.

There's one last elephant in the room: the proteins themselves,

ولكن هناك شيء آخر ايضاً .. انها البروتينات ذاتها

which come from algae, bacteria and funguses

والتي يمكن استخرجها من الطحالب والبكتريا و الفطريات

and all over the tree of life.

ومن جميع اطياف الكائنات الحية

Most of us don't have funguses or algae in our brains,

معظمنا لا يحوي الطحالب او الفطريات في دماغه

ولكن كيف سيتفاعل الدماغ ان قمنا بغرسها فيه

so what will our brain do if we put that in?

هل ستقوم الخلايا بالتجاوب معها .. هل سيهاجمها النظام المناعي

Will the cells tolerate it? Will the immune system react?

ان كل تجاربنا لم تجرب بعد على الانسان

It's early -- these haven't been done in humans yet --

ولكننا نعمل اليوم على كم كبير من الدراسات

but we're working on a variety of studies to examine this.

من اجل تجريب ذلك

وحتى اليوم لم نجد عائقاً او مانعاً للقيام بذلك

So far, we haven't seen overt reactions of any severity

مرتبط بتلك الجزئيات

to these molecules

or to the illumination of the brain with light.

او أعراض جانبية قد تنشأ نتيجة تسليط الضوء على الدماغ

لذا وفي بدايات هذا النطاق الجديد من الابحاث لا اخفيكم اننا متحمسون جداً في خصوصه

So it's early days, to be upfront, but we're excited about it.

I wanted to close with one story,

which we think could potentially be a clinical application.

شيء قد يكون افتراضي

عن طريقة علاج سريرية

Now, there are many forms of blindness

الآن هناك عدة انواع من العمى البصري

بحيث تكون مستقبلات الضوء

where the photoreceptors -- light sensors in the back of our eye --

الحساسة تجاه الموجودة خلف اعيننا معطوبة

are gone.

And the retina is a complex structure.

والشبكية هي نظام معقد

Let's zoom in on it so we can see it in more detail.

ولكن ان استطعنا ان نغوص هنا .. لكي نرى تفاصيل أكثر

خلايا الابصار هي التي نراها هنا على السطح

The photoreceptor cells are shown here at the top.

The signals that are detected by the photoreceptors are transformed

وهناك اشارات يتم التقاطها من قبل خلايا الابصار تلك

ويتم عبر برمجيات معنية تحولها الى نوع آخر من الاشارات

via various computations

حتى في النهاية تصل المعلومات الى خلايا الجيناجليون

until finally, the layer of cells at the bottom, the ganglion cells,

والتي تنقل حزم المعلومات نحو الدماغ

relay the information to the brain,

وما نراه هو إدراك حسي ناتج عن الاشارات تلك

where we see that as perception.

في عدة انواع من العمى البصري مثل التهاب الشبكية الصبغي

In many forms of blindness, like retinitis pigmentosa

or macular degeneration,

والتنكس المعتم

the photoreceptor cells have atrophied or been destroyed.

فان خلايا الابصار تلك تكون معطوبة ..

Now, how could you repair this?

والسؤال هنا .. هل يمكننا اصلاح ذلك

It's not even clear that a drug could cause this to be restored,

حتى اليوم ليس من الواضح ان كان هناك عقار يمكنه القيام بذلك

since there's nothing for the drug to bind to.

لانه لايوجد عقار يمكن معالجة عطب الخلايا البصرية

On the other hand, light can still get into the eye.

ولكن يمكن للضوء ان يخترق العين

لان العين شفافة .. ويمكن للضوء اختراقها

The eye is still transparent and you can get light in.

وان استطعنا ان ناخذ تلك الخلايا البصرية والجزئيات

So what if we could take these channelrhodopsins and other molecules

وقمنا بتركيبها على خلايا رديفة في العين

and install them on some of these other spared cells

يمكننا تحويلهم الى " كميرات " مصغرة

and convert them into little cameras?

ولان العين تحوي العديد من الخلايا

And because there are so many of these cells in the eye,

يمكننا ان نحصل على مجسات بصرية عالية الدقة

potentially, they could be very high-resolution cameras.

انه هذا العمل الذي نعمل عليه

This is some work that we're doing,

ويساعدنا بذلك

led by one of our collaborators, Alan Horsager at USC,

and being sought to be commercialized by a start-up company, Eos Neuroscience,

وقد رأت شركة صاعدة تدعى " اوس نيروسيني " فرصة اقتصادية للاستثمار بذلك

which is funded by the NIH.

What you see here is a mouse trying to solve a six-arm maze.

ان ما ترونه هنا هو فأر يحاول ان يحل معضلة

انها عبارة عن متاهة بستة أذرع .. وهناك القليل من المياه في المتاهة

There's a bit of water to motivate the mouse to move or he'll just sit there.

من اجل تحفيز الفأر على التحرك .. لانه بغير هذا سوف يجلس في مكانه

والهدف من هذه المتاهة

The goal of this maze is to get out of the water

هو حتما الخروج من الماء الى منصة صغيرة

and go to a little platform that's under the lit top port.

تقع في الجهة العليا

ان الفئران ذكية .. ويمكنها ان تحل المتاهة في النهاية

Mice are smart, so this one solves the maze eventually,

but he does a brute-force search.

وهي تقوم ببحث واسع

He's swimming down every avenue until he finally gets to the platform.

فهي تسبح في كل حارة حتى تصل الى المنصة

He's not using vision to do it.

وهلا لا تستخدم بصرها في ذلك

These different mice are different mutations

هذه انواع اخرى من الفئران .. انها طفرات

مصابة بنوع مختلف عن العمى البصري .. الذي يصيب البشر

that recapitulate different kinds of blindness that affect humans.

ونحن ندرس هذه الحالة بصورة مقربة

So we're being careful in trying to look at these different models

لكي نستطيع ان نحصل على تصور عام

so we come up with a generalized approach.

إذاً كيف يمكننا ان نحل ذلك ؟

So how can we solve this?

سنقوم بنفس الاسلوب الذي تحدثت لكم عنه

We'll do exactly what we outlined in the previous slide.

سوف ناخذ تلك المبصرات الضوئية تجاه الضوء الازرق

We'll take these blue light photo sensors

ومن ثم نضعها على طبقة الخلايا

and install them onto a layer of cells

في منتصف الشبكية .. خلف العين

in the middle of the retina in the back of the eye

and convert them into a camera --

لكي نحولهم الى " كميرات " تصوير

just like installing solar cells all over those neurons

بنفس الاسلوب الذي قمنا به بتركيب الخلايا الكهروضوئية على الاعصاب

لكي نجعلها حساسة تجاه الضوء

to make them light-sensitive.

Light is converted to electricity on them.

فالضوء يتحول الى كهرباء

So this mouse was blind a couple weeks before this experiment

وماحدث هو ان الفأر الذي كان مصاب بالعمى البصري قبل هذه التجربة

and received one dose of this photosensitive molecule on a virus.

والذي تلقى جرعة من تلك الجزئيات الكهروضوئية المحملة على فيروس

وكما ترون التطور الحادث هو ان الفأر يستطيع الآن رؤية الجدران

And now you can see, the animal can indeed avoid walls

ومن ثم استطاع الوصول الى المنصة

and go to this little platform

وهاهو الآن يستخدم عينيه عبر المعرفة مرة أخرى

and make cognitive use of its eyes again.

والانجاز الجبار جراء تلك التجربة

And to point out the power of this:

هو ان الفئران المصابة بالعمى استطاعت ان تصل الى تلك المنصة

these animals can get to that platform

just as fast as animals that have seen their entire lives.

بنفس سرعة تلك الفئران المبصرة طيلة حياتها

ان هذه الدراسات ما قبل المخبرية

So this preclinical study, I think, bodes hope

تعطي أملاً كبيراً

for the kinds of things we're hoping to do in the future.

ونحن نأمل في المستقبل

واريد ان اشير الى اننا نستكشف

We're also exploring new business models for this new field of neurotechnology.

مجال تجاري جديد لهذه الادوات التقنية العصبية

نحن نطور هذه الادوات

We're developing tools and sharing them freely

ونتشاركها مع مئات المجموعات حول العالم

with hundreds of groups all over the world

لكي يستطيع الناس دراستها وتجربتها وعلاج الاعتلالات بواسطتها

for them to study and try to treat different disorders.

ونحن نأمل انه وعن طريق اكتشاف الشبكة الدماغية

Our hope is that by figuring out brain circuits

ان نصل الى مرحلة نستطيع فيها تصميم واصلاح تلك الشبكة

at a level of abstraction that lets us repair them and engineer them,

وان نعالج تلك الاعتلالات الدماغية التي اخبرتكم عنها

we can take some of these intractable disorders I mentioned earlier,

والتي حتى اليوم لم نجد لهم علاجاً

practically none of which are cured,

لكي تغدو مجرد ذكرى بعد القضاء عليهم في القرن ال 21

and in the 21st century, make them history.

With blue light, we can drive information, and it's in the form of a one.

And by turning things off, it's more or less a zero.

Our hope is to eventually build brain coprocessors that work with the brain

what neural codes can drive certain behaviors

EB: That's something we're starting to work on very hard.

We're now working on trying to tile the brain with recording elements, too,
