So this is a talk about gene drives,

but I'm going to start by telling you a brief story.

20 years ago, a biologist named Anthony James

بیست سال پیش، یه زیست شناس به نام آنتونی جیمز

got obsessed with the idea of making mosquitos

that didn't transmit malaria.

It was a great idea, and pretty much a complete failure.

فکر فوق‌العاده ای بود، و البته کاملاً شکست خورد.

For one thing, it turned out to be really hard

to make a malaria-resistant mosquito.

James managed it, finally, just a few years ago,

by adding some genes that make it impossible

for the malaria parasite to survive inside the mosquito.

But that just created another problem.

Now that you've got a malaria-resistant mosquito,

how do you get it to replace all the malaria-carrying mosquitos?

There are a couple options,

but plan A was basically to breed up

a bunch of the new genetically-engineered mosquitos

release them into the wild

and hope that they pass on their genes.

The problem was that you'd have to release

literally 10 times the number of native mosquitos to work.

So in a village with 10,000 mosquitos,

you release an extra 100,000.

As you might guess,

this was not a very popular strategy with the villagers.

Then, last January, Anthony James got an email

from a biologist named Ethan Bier.

Bier said that he and his grad student Valentino Gantz

بیر گفت با دانشجوی ارشدش والنتینو گنتز

had stumbled on a tool that could not only guarantee

ابزاری درست کردن که نه تنها تضمین می‌کنه

that a particular genetic trait would be inherited,

که یه صفت ژنتیکی خاص به ارث برسه،

but that it would spread incredibly quickly.

بلکه با سرعت باورنکردنی منتشر بشه.

If they were right, it would basically solve the problem

that he and James had been working on for 20 years.

As a test, they engineered two mosquitos to carry the anti-malaria gene

and also this new tool, a gene drive,

which I'll explain in a minute.

Finally, they set it up so that any mosquitos

that had inherited the anti-malaria gene

wouldn't have the usual white eyes, but would instead have red eyes.

That was pretty much just for convenience

so they could tell just at a glance which was which.

So they took their two anti-malarial, red-eyed mosquitos

and put them in a box with 30 ordinary white-eyed ones,

and let them breed.

In two generations, those had produced 3,800 grandchildren.

That is not the surprising part.

This is the surprising part:

given that you started with just two red-eyed mosquitos

you expect mostly white-eyed descendants.

Instead, when James opened the box,

When I asked Ethan Bier about this moment,

he became so excited that he was literally shouting into the phone.

That's because getting only red-eyed mosquitos

violates a rule that is the absolute cornerstone of biology,

Mendelian genetics.

I'll keep this quick,

but Mendelian genetics says when a male and a female mate,

their baby inherits half of its DNA from each parent.

So if our original mosquito was aa and our new mosquito is aB,

where B is the anti-malarial gene,

the babies should come out in four permutations:

بچه‌هاشون باید به شکل چهار پیش جهش باشن:

aa, aB, aa, Ba.

Instead, with the new gene drive,

they all came out aB.

همه ی اونها aB شدن.

Biologically, that shouldn't even be possible.

So what happened?

پس چه اتفاقی افتاده؟

The first thing that happened

اولین اتفاقی که افتاد

was the arrival of a gene-editing tool known as CRISPR in 2012.

ورود یه ابزار ویرایش ژن به اسم CRISPR در سال ۲۰۱۲ بود.

Many of you have probably heard about CRISPR,

شاید بسیاری از شما درباره ی CRISPR شنیده باشین،

so I'll just say briefly that CRISPR is a tool that allows researchers

پس فقط به طور خلاصه میگم که CRISPR ابزاریه که به محققین اجازه میده

to edit genes very precisely, easily and quickly.

ژنها رو خیلی دقیق، به سادگی و سریع ویرایش کنن.

It does this by harnessing a mechanism that already existed in bacteria.

این کار رو با استفاده از مکانیسمی انجام میده که قبلاً در باکتریها وجود داشت.

Basically, there's a protein that acts like a scissors

اصولاً پروتئینی هست که مثل قیچی عمل میکنه

and cuts the DNA,

و DNA رو میبره،

and there's an RNA molecule that directs the scissors

to any point on the genome you want.

The result is basically a word processor for genes.

You can take an entire gene out, put one in,

میتونین یه ژن کامل رو بردارین، یکی بذارین،

or even edit just a single letter within a gene.

یا حتی یه حرف داخل یه ژن رو ویرایش کنین.

And you can do it in nearly any species.

و میتونین این کار رو تقریباً تو همه گونه‌ها انجام بدین.

OK, remember how I said that gene drives originally had two problems?

خوب، یادتونه که گفتم در ابتدا محرکهای ژنی دو تا مشکل داشت؟

The first was that it was hard to engineer a mosquito

اولی این بود که مشکل می‌شد پشه‌ای رو مهندسی کرد

to be malaria-resistant.

که مقاوم به مالاریا باشه.

That's basically gone now, thanks to CRISPR.

حال این مشکل به کمک CRISPR حل شده.

But the other problem was logistical.

اما مشکل بعدی تدارکاتی بود.

How do you get your trait to spread?

چطور می‌خواین صفت رو گسترش بدین؟

This is where it gets clever.

اینجاست که مشکل پیچیده میشه.

A couple years ago, a biologist at Harvard named Kevin Esvelt

چند سال پیش، زیست شناسی به نام کوین اسولت در هاروارد

wondered what would happen

میخواست ببینه چی میشه

if you made it so that CRISPR inserted not only your new gene

اگه کاری کنین که CRISPR علاوه بر ژنهای جدیدتون

but also the machinery that does the cutting and pasting.

ماشینی رو وارد کنه که عمل برش و چسبوندن رو هم انجام بده؟

In other words, what if CRISPR also copied and pasted itself.

به عبارت دیگه، چی میشه اگه CRISPR خودش کپی پیست کنه؟

You'd end up with a perpetual motion machine for gene editing.

حاصلش یه ماشین در حرکت دائمیه برای ویرایش ژن.

And that's exactly what happened.

و این دقیقاً همون اتفاقیه که افتاد.

This CRISPR gene drive that Esvelt created

محرک ژن CRISPR که اسولت ساخت

not only guarantees that a trait will get passed on,

علاوه بر تضمین انتقال یه صفت،

but if it's used in the germline cells,

اگه در سلول های رده زایا استفاده بشه،

it will automatically copy and paste your new gene

به طور خودکار ژنهای جدید رو کپی پیست میکنه

into both chromosomes of every single individual.

به داخل هر دو کروموزوم تک تک افراد.

It's like a global search and replace,

این مثل یه جستجو و جایگزینی سراسریه،

or in science terms, it makes a heterozygous trait homozygous.

یا به اصطلاح علمی، یه صفت هتروزیگوت رو هموزیگوت میکنه.

So, what does this mean?

خوب، معنی این چیه؟

For one thing, it means we have a very powerful,

به بیانی، معنیش اینه که ما ابزاری بسیار قدرتمند

but also somewhat alarming new tool.

اما تا حدودی هشدار دهنده داریم.

Up until now, the fact that gene drives didn't work very well

تا این لحظه این واقعیت که محرک ژن خوب کار نمی کنه

was actually kind of a relief.

در واقع خیال همه رو راحت کرده.

Normally when we mess around with an organism's genes,

در حالت طبیعی وقتی روی ژنهای جانداری خرابکاری می‌کنیم،

we make that thing less evolutionarily fit.

باعث میشیم از نظر فرگشتی نزول کنه.

So biologists can make all the mutant fruit flies they want

یعنی زیست شناسان میتونن هر نوع مگس میوه جهش یافته‌ای که میخوان درست کنن

without worrying about it.

بدون اینکه نگرانش باشن.

If some escape, natural selection just takes care of them.

اگه بعضی از اونا رها بشن، انتخاب طبیعی مراقبشون هست.

What's remarkable and powerful and frightening about gene drives

چیزی که درباره ی محرکهای ژنی قابل توجه و قدرتمند و نگران کننده است

is that that will no longer be true.

اینه که نکته فوق دیگه درست نباشه.

Assuming that your trait does not have a big evolutionary handicap,

با این فرض که صفت شما دچار نقیصه ی فرگشتی بزرگی نباشه،

like a mosquito that can't fly,

مثل پشه ای که نتونه پرواز کنه،

the CRISPR-based gene drive will spread the change relentlessly

محرک ژنی مبتنی بر CRISPR به طور مداوم تغییر رو گسترش میده

until it is in every single individual in the population.

تا وقتی به تک تک افراد اون جمعیت منتقل بشه.

Now, it isn't easy to make a gene drive that works that well,

البته ساختن ابزاری که به این خوبی کار کنه آسون نیست،

but James and Esvelt think that we can.

اما به نظر جیمز و اسولت ما میتونیم.

The good news is that this opens the door to some remarkable things.

خبر خوب اینه که این ابزار راه رو برای چیزهای جالبی باز میکنه.

If you put an anti-malarial gene drive

اگه یه محرک ژن ضدمالاریا رو

in just 1 percent of Anopheles mosquitoes,

فقط به ۱ درصد پشه های آنوفل وارد کنیم،

the species that transmits malaria,

گونه ای که مالاریا رو منتقل میکنه،

researchers estimate that it would spread to the entire population in a year.

محققین تخمین میزنن در عرض یک سال این بین همه ی جمعیت پخش میشه.

So in a year, you could virtually eliminate malaria.

یعنی در عرض یک سال میتونین مالاریا رو تقریباً ریشه کن کنین.

In practice, we're still a few years out from being able to do that,

در عمل، هنوز چند سال مونده تا بتونیم این کار رو بکنیم،

but still, a 1,000 children a day die of malaria.

اما همچنان هر روز هزار کودک از مالاریا میمیره.

In a year, that number could be almost zero.

این تعداد میتونه در عرض یک سال تقریباً صفر بشه.

The same goes for dengue fever, chikungunya, yellow fever.

همین در مورد تب دنگی، چیکونگونیا، و تب زرد هم صدق میکنه.

And it gets better.

و بهتر میشه.

Say you want to get rid of an invasive species,

مثلاً میخواین از شر گونه های مهاجم خلاص بشین.

like get Asian carp out of the Great Lakes.

مثلاً ماهی کپور آسیایی رو از آبهای گریت لیکز ریشه کن کنین.

All you have to do is release a gene drive

تنها کاری که باید بکنین رها کردن یه محرک ژنیه،

that makes the fish produce only male offspring.

که باعث بشه ماهی ها فقط بچه های نر تولید کنن.

In a few generations, there'll be no females left, no more carp.

بعد از چند نسل، هیچ ماهی ماده ای و هیچ کپوری باقی نمی‌مونه.

In theory, this means we could restore hundreds of native species

مفهوم نظری اش اینه که میتونیم صدها گونه ی بومی رو برگردونیم

that have been pushed to the brink.

که در معرض انقراض هستن.

OK, that's the good news,

خوب، این خبر خوب بود.

this is the bad news.

خبر بد اینه که

Gene drives are so effective

محرکهای ژن اونقدر قوی هستن که

that even an accidental release could change an entire species,

حتی رهاسازی تصادفی ممکنه یه گونه رو به طور کلی،

and often very quickly.

و گاهی به سرعت عوض کنه.

Anthony James took good precautions.

آنتونی جیمز خیلی احتیاط کرد.

He bred his mosquitos in a bio-containment lab

او جفت گیری پشه ها رو داخل یه آزمایشگاه ایمن انجام داد

and he also used a species that's not native to the US

و همچنین از گونه ای استفاده کرد که بومی آمریکا نبود

so that even if some did escape,

تا حتی اگه تعدادی رها بشه،

they'd just die off, there'd be nothing for them to mate with.

نابود بشه و چیزی نباشه که باهاش جفت گیری کنه.

But it's also true that if a dozen Asian carp with the all-male gene drive

accidentally got carried from the Great Lakes back to Asia,

they could potentially wipe out the native Asian carp population.

And that's not so unlikely, given how connected our world is.

In fact, it's why we have an invasive species problem.

And that's fish.

Things like mosquitos and fruit flies,

there's literally no way to contain them.

They cross borders and oceans all the time.

OK, the other piece of bad news

خوب، قسمت دیگه ی خبر بد

is that a gene drive might not stay confined

اینه که یه محرک ژنی ممکنه محدود باقی نمونه

to what we call the target species.

به چیزی که ما میگیم گونه های هدف.

That's because of gene flow,

علتش جریان ژنه،

which is a fancy way of saying that neighboring species

که توضیح ساده‌ش اینه که گونه های همسایه

sometimes interbreed.

گاهی با هم جفت گیری می‌کنن.

If that happens, it's possible a gene drive could cross over,

اگه این اتفاق بیفته، ممکنه یه محرک ژنی به طور متقاطع منتقل بشه،

like Asian carp could infect some other kind of carp.

مثلاً کپور آسیایی میتونه یه نوع کپور دیگه رو آلوده کنه.

That's not so bad if your drive just promotes a trait, like eye color.

اگه این محرک محدود به یه صفت، مثل رنگ چشم باشه، خیلی بد نیست.

In fact, there's a decent chance that we'll see

در واقع، شانس خوبی داریم که

a wave of very weird fruit flies in the near future.

در آینده نزدیک انواع رنگارنگ مگس های میوه رو در طبیعت ببینیم.

But it could be a disaster

اما این میتونه یه فاجعه باشه

if your drive is deigned to eliminate the species entirely.

اگه محرک طوری طراحی شده باشه که یه گونه رو به طور کلی منقرض کنه.

The last worrisome thing is that the technology to do this,

آخرین نگرانی اینه که فناوری انجام این کار،

to genetically engineer an organism and include a gene drive,

is something that basically any lab in the world can do.

An undergraduate can do it.

A talented high schooler with some equipment can do it.

Now, I'm guessing that this sounds terrifying.

Interestingly though, nearly every scientist I talk to

seemed to think that gene drives were not actually that frightening or dangerous.

Partly because they believe that scientists will be

very cautious and responsible about using them.

So far, that's been true.

But gene drives also have some actual limitations.

So for one thing, they work only in sexually reproducing species.

So thank goodness, they can't be used to engineer viruses or bacteria.

پس خوشبختانه نمیشه از اون برای مهندسی ویروس ها یا باکتری ها استفاده کرد.

Also, the trait spreads only with each successive generation.

همچنین، گسترش یه صفت فقط از نسلی به نسل بعد انجام میشه.

So changing or eliminating a population

پس تغییر یه جمعیت یا حذف اون

is practical only if that species has a fast reproductive cycle,

تنها در صورتی عملی است که گونه ی مورد نظر چرخه تولیدمثلی سریعی داشته باشه،

like insects or maybe small vertebrates like mice or fish.

مثل حشرات یا شاید مهره داران کوچکی مثل موش یا ماهی.

In elephants or people, it would take centuries

در فیل یا انسان، ممکنه قرن ها طول بکشه

for a trait to spread widely enough to matter.

تا یه صفت به حدی گسترش پیدا کنه که اهمیت داشته باشه.

Also, even with CRISPR, it's not that easy to engineer a truly devastating trait.

همچنین، حتی با CRISPR، مهندسی یه صفت ویرانگر اونقدر هم ساده نیست.

Say you wanted to make a fruit fly

مثلاً اگه بخواین مگس میوه‌ای تولید کنین

that feeds on ordinary fruit instead of rotting fruit,

که به جای میوه فاسد از میوه سالم تغذیه کنه،

with the aim of sabotaging American agriculture.

با هدف انهدام کشاورزی آمریکا.

First, you'd have to figure out

اول باید بفهمین

which genes control what the fly wants to eat,

which is already a very long and complicated project.

که خودش یه پروژه ی خیلی طولانی و پیچیده س.

Then you'd have to alter those genes to change the fly's behavior

to whatever you'd want it to be,

which is an even longer and more complicated project.

که این حتی پروژه ی طولانی‌تر و پیچیده‌تری است.

And it might not even work,

because the genes that control behavior are complex.

چون ژنهایی که رفتار رو کنترل میکنن پیچیده هستن.

So if you're a terrorist and have to choose

پس اگه شما یه تروریست باشین و بخواین انتخاب کنین

between starting a grueling basic research program

بین شروع یه برنامه‌ی تحقیقاتی خسته کننده

that will require years of meticulous lab work and still might not pan out,

که نیاز به سالها کار دقیق آزمایشگاهی داره و حتی ممکنه موفق نشه،

or just blowing stuff up?

یا این که فقط چیزهایی رو منفجر کنین؟

You'll probably choose the later.

احتمالاً دومی رو انتخاب می‌کنین.

This is especially true because at least in theory,

it should be pretty easy to build what's called a reversal drive.

ساختن چیزی که بهش میگن محرک معکوس باید خیلی آسون باشه.

That's one that basically overwrites the change made by the first gene drive.

So if you don't like the effects of a change,

you can just release a second drive that will cancel it out,

at least in theory.

OK, so where does this leave us?

We now have the ability to change entire species at will.

Should we?

باید تغییر بدیم؟

Are we gods now?

آیا اکنون خدا هستیم؟

I'm not sure I'd say that.

مطمئن نیستم اینو بخوام بگم.

But I would say this:

first, some very smart people

اول، برخی افراد خیلی باهوش

are even now debating how to regulate gene drives.

حتی حالا دارن درباره ی مقررات تنظیم محرکهای ژنی بحث میکنن.

At the same time, some other very smart people

به طور همزمان، برخی افراد خیلی باهوش دیگه

are working hard to create safeguards,

به سختی دارن کار می کنن تا ابزارهای حفاظتی ایجاد کنن،

like gene drives that self-regulate or peter out after a few generations.

That's great.

But this technology still requires a conversation.

And given the nature of gene drives,

that conversation has to be global.

What if Kenya wants to use a drive but Tanzania doesn't?

Who decides whether to release a gene drive that can fly?

کی تصمیم میگیره محرک ژنی ساخته بشه که بتونه پرواز کنه؟

I don't have the answer to that question.

من جوابی واسه این سؤال ندارم.

All we can do going forward, I think,

By that I mean, not just the choice to use a gene drive,

منظورم از انتخاب فقط این نیست که از یه محرک ژنی استفاده کنیم،

but also the choice not to use one.

بلکه این هم هست که از یه محرک ژنی استفاده نکنیم.

Humans have a tendency to assume that the safest option

بشر تمایل داره به این که فرض کنه بی‌خطرترین گزینه

is to preserve the status quo.

حفظ وضعیت موجوده.

But that's not always the case.

اما این همیشه درست نیست.

Gene drives have risks, and those need to be discussed,

محرکهای ژنی خطرهایی دارن، و باید درباره‌ش بحث بشه،

but malaria exists now and kills 1,000 people a day.

اما در حال حاضر مالاریا وجود داره و هر روز یک هزار نفر رو میکشه.

To combat it, we spray pesticides that do grave damage to other species,

ما برای مبارزه با اون، از حشره‌کش‌هایی استفاده می‌کنیم که به سایر گونه‌ها آسیب می‌زنه،

including amphibians and birds.

So when you hear about gene drives in the coming months,

and trust me, you will be hearing about them,

It can be frightening to act,
