It's a devastating disease that takes an enormous emotional toll.

Not only on the patient, but the patient's loved ones, as well.

It is a battle that the human race has been fighting for centuries.

がんとの戦いは数世紀に渡って 人類が続けてきた闘争です

And while we've made some advancements,

we still haven't beaten it.

未だに打ち負かす事は 出来ずにいます

Two out of five people in the US will develop cancer in their lifetime.

アメリカでは５人に２人が 生涯の内で がんに罹ります

Of those, 90 percent will succumb to the disease due to metastases.

その内 90％の人達が転移の為に がんで亡くなっています

Metastasis is a spread of cancer from a primary site to a distal site,

転移とは 血液循環や リンパ系を通した

through the circulatory or the lymphatic system.

原発から遠隔までの がんの広がりの事です

For instance, a female patient with breast cancer

例えば 乳がんの女性患者は

doesn't succumb to the disease simply because she has a mass on her breast.

単に胸に出来た腫瘤で 亡くなるのではありません

She succumbs to the disease because it spreads

がんが肺、肝臓、リンパ節、脳、骨 に広がり

to the lungs, liver, lymph nodes, brain, bone,

そこでがんは障害を起こし

where it becomes unresectable or untreatable.

治療も出来ずに 亡くなるのです

Metastasis is a complicated process.

転移とは 複雑な過程です

One that I've studied for several years now.

これまで 数年に渡って研究を行ってきました

And something that my team and I discovered recently

我がチームが 最近発見したのは

was that cancer cells are able to communicate with each other

がん細胞は お互いに情報を交換し

and coordinate their movement,

自分達がどれくらい密に腫瘍の中に

based on how closely packed they are in the tumor microenvironment.

詰め込まれているかを基に その動きを 協調させている事です

They communicate with each other through two signaling molecules

がん細胞は２つのシグナル分子― インターロイキン６と８を通して

called Interleukin-6 and Interleukin-8.

情報の伝達を行っています

Now, like anything else in nature,

さて 自然の中の 他の全てのものと同様

when things get a little too tight, the signal is enhanced,

窮屈になると 信号が増強され

causing the cancer cells to move away faster from the primary site

がん細胞は 原発部位から離れて 新しい部位に広がる速度を速める

and spread to a new site.

原因となるのです

So, if we block this signal, using a drug cocktail that we developed,

そこで この信号を私達が開発した カクテル（薬の組み合わせ）を使って阻止すれば

we can stop the communication between cancer cells

がん細胞同士の情報伝達を阻止し

and slow down the spread of cancer.

がんの広がりを 遅くする事が出来るのです

Let me pause here for a second

ちょっと ここで一息ついて

and take you back to when this all began for me in 2010,

私にとって 全てが始まった 2010年に遡りましょう

when I was just a sophomore in college.

私は大学２年生でした

I had just started working in Dr Danny Wirtz's lab

ジョンズ・ホプキンス大学の ダニー・ワーツ博士の研究室で

at Johns Hopkins University.

研究を始めたばかりでした

And I'll be honest: I was a young, naive, Sri Lankan girl,

正直言って 私はまだ若くて うぶな スリランカの女の子で

who had no previous research experience.

研究の経験は 全くありませんでした

And I was tasked to look at how cancer cells move

がん細胞が 培養皿の中に再現された

in a 3D collagen I matrix

that recapsulated, in a dish,

the conditions that cancer cells are exposed to in our bodies.

これはがん細胞が体内で 曝されているのと同じ条件です

This was new and exciting for me,

because previous work had been done on 2D, flat, plastic dishes

that really weren't representative

of what the cancer cells are exposed to in our bodies.

Because, let's face it,

the cancer cells in our bodies aren't stuck onto plastic dishes.

体内のがん細胞は プラスチックの 培養皿の上になど 乗ってはいません

It was during this time that I attended a seminar

その頃 プリンストン大学の

conducted by Dr Bonnie Bassler from Princeton University,

ボニー・バスラー博士のセミナーに 出席しました

where she talked about how bacteria cells communicate with each other,

そこで彼女は 細菌が その密度に基づいて どのように

based on their population density, and perform a specific action.

互いに情報伝達をし 特定の行動を とるかについて話をしました

It was at this moment that a light bulb went off in my head, and I thought,

頭の中の電球が閃いた この瞬間 私はこう思いました

"Wow, I see this in my cancer cells every day,

「あぁ 動きと言えば 私は毎日 がん細胞の動きを

when it comes to their movement."

観察しているわ」

The idea for my project was thus born.

私のプロジェクトのアイディアは こうして生まれました

I hypothesized that cancer cells are able to communicate with each other

がん細胞は 腫瘍の微小環境の中で どれ程 密に詰まっているかに基づき

and coordinate their movement,

互いに情報伝達ができ

based on how closely packed they are in the tumor microenvironment.

その動きを協調させるのだという 仮説を立てました

I became obsessed with pursuing this hypothesis.

私はこの仮説を追求する事に 取り憑かれました

And fortunately, I work for someone

そして運良く この奇抜な考えに 付き合ってくれる

who is open to running with my crazy ideas.

余裕がある人と 研究をしています

So, I threw myself into this project.

そこで私は このプロジェクトに没頭しました

However, I couldn't do it by myself.

しかしながら この研究は 自分１人では出来ませんでした

I needed help.

私には助けがー

I definitely needed help.

絶対的に助けが 必要だったのです

So we recruited undergraduate students, graduate students,

そこで 私が大学２年生で思いついた このアイディアに

postdoctoral fellows and professors

一緒に取り組んでくれる 異なる施設や

from different institutions and multiple disciplines

多様な学科からの 大学生や大学院生

to come together and work on this idea

ポストドク研究員、教授に

that I conceived as a sophomore in college.

私達は協力を募りました

After years of conducting experiments together

彼らと共に 数年実験を行い

and merging different ideas and perspectives,

異なる概念や観点を 融合させた後

we discovered a new signaling pathway

がん細胞がどうやって互いに 密度を基に

that controls how cancer cells communicate with each other and move,

情報伝達を行い 移動するかを制御する 新たなシグナル伝達経路を

based on their cell density.

私達は発見したのです

Some of you might have heard this,

耳にした方が いるかも知れません

because most of social media knows it as the Hasini effect.

SNSではこれを「ハシニ効果」と 呼んでいますから

And we weren't done yet.

それで終わりではありませんでした

We then decided that we wanted to block this signaling pathway

その後 私達はこのシグナル伝達経路を ブロックし がんの広がる速度を

and see if we could slow down the spread of cancer.

落とすことが出来るかどうかを 確かめる事にしました

Which we did, in preclinical animal models.

私達はこれを 臨床試験前の動物実験として行いました

We came up with a drug cocktail consisting of tocilizumab,

私達はトシリズマブを含めた カクテルを思いつきました

which is currently used to treat rheumatoid arthritis,

これは現在 関節リウマチの 治療に使われています

and reparixin, which is currently in clinical trials against breast cancer.

レパリキシンは 現在乳がんに対する 治験が行われています

And interestingly, what we found was that this cocktail of drugs

又 興味深い事に このカクテルは

really had no effect on tumor growth,

実際 腫瘍の増大には 何の効果もないものの

but directly targeted metastases.

転移を直接 標的にする事が 分かりました

This was a significant finding,

これは重要な発見でした

because currently, there aren't any FDA-approved therapeutics

現在 直接がんの広がりを 標的にする

that directly target the spread of cancer.

FDAが承認した治療薬は ないからです

In fact, the spread of cancer, metastasis,

実際 がんの広がり―転移は

is thought of as a byproduct of tumor growth.

腫瘍増大の副産物と 考えられているのです

Where the idea is, if we can stop the tumor from growing,

そこで 腫瘍の増大を 食い止められるなら

we can stop the tumor from spreading.

腫瘍の広がりも止められると 考える訳です

However, most of us know that this is not true.

しかしながら これは 事実ではないと分かっています

We, on the other hand, came up with the drug cocktail

一方 私達は腫瘍の増大を 標的にする事によってではなく

that targets metastasis not by targeting tumor growth,

それを支配する複雑なメカニズムを 標的にする事によって

but by targeting the complex mechanisms that govern it,

転移を標的にする薬のカクテルを 思いついたのです

through the targeting of the Hasini effect.

ハシニ効果を標的にして―

This work was recently published in "Nature Communications,"

この研究は最近『Nature Communications』に掲載され

and my team and I received an overwhelming response from around the world.

私達のチームは 世界中から 圧倒的な反響を受けています

Nobody on my team could have predicted this sort of response.

チームの内誰も こんな反応があるとは 予想していませんでした

We seem to have struck a nerve.

多くの研究者の心をとらえたのです

Looking back, I am extremely grateful for the positive response that I received,

振り返ると 私は学者からだけでなく この恐ろしい病気に冒された

not only from academia, but also patients,

世界中の患者達からの支持を

and people around the world affected by this terrible disease.

大変有難いと思っています

As I reflect on this success I've encountered with the Hasini effect,

ハシニ効果との出会いという この成功を思い起こすと

I keep coming back to the people that I was fortunate enough to work with.

幸運にも 共同研究できた人達が 常に思い出されます

The undergraduate students

その勤労と献身とを通して

who demonstrated superhuman powers through their hard work and dedication.

超人的な力を見せてくれた大学生達

The graduate students and the postdoctoral fellows,

大学院生やポストドク研究員

my fellow Avengers, who taught me new techniques

私に新たな技術を教え 仕事が順調に進むようにしてくれた

and always made sure I stayed on track.

The professors, my Yodas and my Obi-Wan Kenobis,

who brought their expertise into making this work into what it is today.

and never let us give up on our ambitious endeavors.

It took a village to help me study metastasis.

私が転移の研究をする手助けを してくれたのは共同体でした

And believe me, without my village, I wouldn't be here.

共同体がなければ 絶対に 私はここまで来れなかったでしょう

Today, our team has grown,

こんにち 私達のチームは 成長を遂げ

and we are using the Hasini effect to develop combination therapies

腫瘍の増大と転移を 効果的に標的にする

that will effectively target tumor growth and metastases.

組み合わせ治療の開発に ハシニ効果を利用しています

We are engineering new anticancer therapeutics,

私達は 薬物毒性を抑え 薬剤耐性を減らす

to limit toxicity and to reduce drug resistance.

新たな抗がん治療薬を 作ろうとしています

And we are developing groundbreaking systems

更により良いヒトへの臨床治験 開発の手助けとなる

that will help for the development of better human clinical trials.

画期的なシステムを 開発しています

It blows my mind to think that all this,

現在自分が探求している 想像を超えた研究の事や

the incredible work that I'm pursuing --

今日このステージに立って

and the fact that I'm standing here, talking to you today --

皆さんにお話ししていることが

all came from this tiny idea

全て 私がまだ20歳の時

that I had when I was sitting at the back of a seminar

セミナーの後ろの席で思いついた 些細なアイディアから

when I was just 20 years old.

生まれたと考えると 驚きを禁じえません

I recognize that right now, I am on this incredible journey

私は今まさに 大きな情熱を傾ける研究に邁進でき

that allows me to pursue work that I am extremely passionate about,

毎日 好奇心を 掻き立てられる

and something that feeds my curiosity on a daily basis.

驚くべき道のりの 途中にいます

But I have to say, my favorite part of all of this --

ただ 間違いなく 私が一番気に入っていることは—

other than, of course, being here, talking to you, today --

勿論 皆さんにここで お話しできることを除いてですが—

is the fact that I get to work with a diverse group of people,

研究をより確固たるものにし 何にもましてずっと面白くしてくれた

who make my work stronger, better and just so much more fun.

多様な人達のグループと共に 私は研究をしてきたという事実です

And because of this, I have to say

そこで こう言わざるを得ません

that collaboration is my favorite superhuman power.

協力とは 私が大好きな 超人的な力です

And what I love about this power is that it's not unique to me.

この力を 私が気に入っているのは それが 自分だけに備わったものではなく

It's within all of us.

私達皆の中にあるものだ という事です

My work shows that even cancer cells

がん細胞でさえ 人間の身体を侵し 被害を広げる為には

use collaboration to invade our bodies and spread their wrath.

協力し合うものだと 私の研究で 明らかになりました

For us humans, it is a superpower that has produced incredible discoveries

私達人間にとって 医学的 科学的な分野で 途方もない発見を

in the medical and scientific field.

生み出してきたのは この超人的な力です

And it is the superpower that we can all turn to

私達皆が 自分自身よりも 大きなものを創造し

to inspire us to create something bigger than ourselves,

世界をより良いものにする 手助けとなるよう 促す為に

that will help make the world a better place.

頼りに出来るものは この超人的な力なのです

Collaboration is the superpower that I turn to, to help me fight cancer.

協力とは 私ががんと闘う手助けとなり 頼れる超人的な力なのです

And I am confident that with the right collaborations,

適切な協力があれば がんに打ち勝つのだと

we will beat this terrible disease.

私は確信しています
